iTunes is, without question, the Daddy.
SCurrently, the shows I run and work with tend to show around 60% to 80% of all traffic coming from iTunes alone. Some Podcasts see 90%+ of their traffic from iTunes, but that probably means they’re not offering it up many other places. However we dice it, though, and however we promote shows, iTunes is always our biggest traffic source by at least a factor of 10x. Eg. iTunes is showing 65% and the next best single source is 6.5% max.

How Long Does it Take to Get In?
This is a ‘how long is a piece of string’ question I’m afraid. I’ve had shows approved in under 2 hours, but I’ve also had shows that took more than 3 days to get approval.
My recommendation is to allow 3 working days at a minimum. It’s very unlikely it’ll take longer than that. Plan your iTunes submission to be at least that many weekdays in front of your advertised launch.
And, in fact, if you’re advertising launch dates, and relying on it being live by then, allow 5 working days, just in case.
